In situ expression of Th17 immunologic mediators in American cutaneous leishmaniasis caused by Leishmania (V.) braziliensis and Leishmania (L.) amazonensis in the Brazilian Amazon

American cutaneous leishmaniasis (ACL) presents a wide spectrum of clinical and immunopathological manifestations. In Brazil, Leishmania (L.) amazonensis [La] Leishmania(V.)braziliensis [Lb] show the highest pathogenic potential for humans causing different forms: localized (LCL : Lb/La), anergic diffuse (ADCL La) mucocutaneous (MCL Lb). ADCL MCL are most severe forms infection leads to cellular immune response at hyposensitivity hypersensitivity poles. Th17-cells involved in ACL pathogenesis, derived from naïve TCD4+ cells regulated by RORγt, differentiate presence IL-6, TGF-β, IL- 1β, IL-23 express IL-17. Aim this study was characterize mediated Th17-profile through situ determination expression IL-17, IL-1β, clinical-immunopathological caused L.(L.)amazonensis L.(V.)braziliensis . Biopsies skin mucosal lesions forty patients including ADCL(n=8), LCL[La](n=17), LCL[Lb](n=9) MCL(n=6), were examined immunohistochemistry. The immunostained density (cells/mm 2 ) determined image analysis system using AxionVision 4.8 software (Zeiss). As disease evolution time (DET) among patients, effect DET on immunological markers evaluated histopathological changes, ANCOVA. Our results showed significantly increased IL-1β with polar MCL); higher TGF-β found ADCL. influenced RORγt IL-6 in: categories parasitism. also affected production types inflammatory infiltrate, evidencing that had Th17 profile cytokines ACL. Together, mediators spectrum, as well effect, demonstrate participation cell lineage immunopathogenesis ACL, mainly more spectrum. dubious role played may favors suppression parasitic persistence ADCL, while it contributes an exacerbated parasite scarcity.
